CJC-1295 without DAC, also referred to as Modified GRF (1-29) or Mod GRF 1-29, is a tetrasubstituted analogue of the first 29 amino acids of endogenous GHRH. The four amino acid substitutions confer enhanced enzymatic stability while preserving GHRH receptor binding affinity, yielding an approximate plasma half-life of 30 minutes in research models.

Unlike the DAC-conjugated version, CJC-1295 without DAC produces discrete, short-duration GH pulses that more closely approximate physiological GH secretion patterns observed in vivo. This characteristic makes it a preferred model compound for studying pulse-mode somatotropic stimulation and for research protocols that require temporal control of GH axis activation.

Research applications include acute GH secretion studies, circadian GH rhythm investigations, and synergistic protocols with GHRP compounds including Ipamorelin. Studies have characterised its binding kinetics at the GHRH receptor, signal transduction pathways involving cAMP and protein kinase A, and the downstream effects on pituitary somatotroph cell biology.
