MOTS-C (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a mitochondria-derived peptide encoded within the 12S ribosomal RNA gene of the mitochondrial genome. Research has identified it as a key regulator of insulin sensitivity and glucose metabolism, with preclinical studies demonstrating its ability to activate AMPK signalling pathways and modulate the AICAR/AMPK/mTOR axis.

In animal model research, MOTS-C has been associated with improvements in metabolic flexibility, enhanced exercise performance, and lifespan extension under high-fat diet conditions. Its dual role as both a mitochondrial and nuclear signalling molecule makes it a unique subject in longevity and metabolic research — one of very few peptides known to originate from the mitochondrial genome.

Research has also explored MOTS-C in the context of age-related insulin resistance, adipogenesis inhibition, and skeletal muscle glucose uptake modulation. Its emerging role as a “mitokine” — a mitochondria-derived hormone-like signalling peptide — positions it at the frontier of contemporary bioenergetics and longevity research.
